Thank you for the further information. It is difficult to select alternatives that are the most 'specific' as we have not tested these antibodies side by side in the same sample application or species. Reviewing the datasheets I think that ab74906 anti-Rab7 antibody and ab3612 anti-Rabb11 antibody may be preferable but please do check the datasheets to confirm whether you agree. Please bear in mind that ab74906 (rabbit polyclonal to Rab7) is not tested in ICC, and as it shows a weak band in WB the staining intensity may be lower than other antibodies. In addition, ab3612 (rabbit polyclonal to Rab11) is only predicted to work in mouse samples, and may not give positive results. As the Rab7 and Rab11 antibodies will be used in previously untested species/applications, would it be possible to submit an Abreview with your results? It doesn't matter if it is positive or negative, but the information will be really useful for other researchers that may be interested in using these products in a similar manner. I will be able to provide ab78970 and ab87802. Could you also submit Abreview information for these products too? ICC/IF image of ab50533 stained HepG2 cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab50533, 1µg/ml) overnight at +4ºC. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
