The RAB14 protein has several Ser/Thr residues which may be phosphorylated (SwissProt data). This could cause the doublet observed at the expected molecular weight.
ab40938 staining RAB14 in human HeLa cells by Immunocytochemistry/ Immunofluorescence. HeLa cells were infected with C. trachomatis serovar L2 (MOI 5) for 10 hours. The cells were fixed in paraformaldehyde, permeabilised with saponin and then blocked using 0.2% BSA for 1 hour at 25ºC. Samples were then incubated with primary antibody at 1/100 for 1 hour at 25ºC. The secondary antibody used was a goat anti-rabbit IgG conjugated to FITC (green) used at a 1/200 dilution. Microtubules were evinced by a mouse monoclonal anti-ß-tubulin (1/200) followed by donkey anti-mouse Cy5-labeled antibodies (1/700) (red). Endogenous Rab14 was recruited to the chlamydial inclusion (blue asterisk). Bacteria were labeled with Hoechst 0.1 mg/ml (blue). ab40938 was useful to detect for the first time the recruitment of an endogenous RAB to the chlamydial inclusion.
