Rab 25 is a plasma membrane-associated small GTPase which cycles between an active GTP-bound and an inactive GDP-bound state. Rab proteins play an important role for either in endocytosis or in biosynthetic protein transport. The possibility that Rab proteins might also direct the exocytosis from secretory vesicles to the plasma membrane is supported by the observation that in yeast, the SEC4 protein, which is 40% similar to Rab proteins, is associated with secretory vesicles. Rab proteins are located on the cytoplasmic face of organelles and vesicles and are involved in intracellular membrane fusion reactions. Rab25 was cloned from a gastric parietal cell cDNA library and is expressed in epithelial tissues such as the gastrointestinal mucosae, kidney, and lung.

Immunocytochemistry/ Immunofluorescence - Rab25 antibody (ab45855)
ICC/IF image of ab45855 stained HeLa cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab45855, 5µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM. This antibody also gave a positive result in 100% methanol fixed (5 min) Hek293 cells at 5µg/ml, and in 4% PFA fixed (10 min) HeLa and MCF7 cells at 5µg/ml.
