The lysates were aliquoted and kept in -80?C. AMOUNT OF PROTEIN LOADED 50ug-150ug ELECTROPHORESIS/GEL CONDITIONS 10-12% reducing gel TRANSFER AND BLOCKING CONDITIONS 60 min up to 90 min wet transfer containing 20% methanol. Blocking with 5% milk in PBS 60 min. SECONDARY ANTIBODY anti mouse HRP conjugated 1:3000 to 1:10000 HOW MANY TIMES HAVE YOU TRIED THE APPLICATION? 6 HAVE YOU RUN A "NO PRIMARY" CONTROL? Yes DO YOU OBTAIN THE SAME RESULTS EVERY TIME? Yes WHAT STEPS HAVE YOU ALTERED? None ADDITIONAL NOTES The protcol described here has been used for at least 10 other antibodies among them several are from abcam but we have problems only with the Rab11A ab11034 antibody. I am absolutely sure that there is nothing wrong with the protocol. I would really appreciate if you could me send an answer as soon as possible otherwise I will have to order this from another company and finish this last experiment for our manuscript. It is quite urgent.
