Rab3A is a 25kD GTP-binding protein implicated in neurotransmitter release. The geranylgeranyl groups at the carboxyl-terminus of Rab3A are essential for its association with synaptic vesicles, guanine nucleotide-dissociation factor (GDI), and for GTP binding. Rab3A is believed to regulate vesicle fusion through a cycle of GDP/GTP exchange and GTP hydrolysis, which is in turn modulated by GTPase activating protein (GAP), GDI, guanine nucleotide-releasing factor (GRF), and rabphilin. The dif-ferent guanine nucleotide binding states of Rab3A is postulated to affect its ability to associate or dissociate with membranes during synaptic vesicle recycling. Its GTP-bound form, which represents the active form of Rab3A, associates with membrane and regulates vesicle docking and fusion. Rab3A and its homolog, Rab3C, have been shown to be co-localized to and dissoci-ated from synaptic vesicles after the stimulation of exocytosis. Deletion of the Rab3A structural gene has minimal effect on the morphology of the nervous system and spontaneous and evoked synaptic currents. However, Rab3A knockout mice exhibited that an increased rate of synaptic depression after repeated stimulation, suggesting Rab3A is involved in maintaining a steady supply of vesicles for release.
Rab proteins are low-molecular-weight GTP-binding proteins that form the largest branch of the Ras superfamily of GTPases. Located on the cytoplasmic face of organelles and vesicles, rab proteins are involved in intracellular membrane fusion reactions. Three membrane proteins, synaptosomal associated protein of 25 kDa (SNAP-25), synaptobrevin and syntaxin, form the core of a ubiquitous membrane fusion machine that interacts with the soluble proteins N-ethylmaleimide-sensitive factor (NSF) and a-SNAP.
Rab proteins, in co-ordination with the core fusion machinery and Munc-18, help to mediate vesicle docking and fusion. There exist over 40 rab proteins that have been described in mammals and the best studied is rab 3A. Rab 3A is found to be abundant in presynaptic nerve terminals and is also found to be crucial in acrosomal exocytosis in human spermatozoa.
Abnormal accumulation of rab 3A in the cytoplasm of Purkinje cells has been reported in the prion protein related Creutzfeldt-Jakob disease.

Immunocytochemistry/ Immunofluorescence - Rab3A antibody (ab3335)
ab3335 staining hamster CHO-K1 cells by ICC/IF. Cells were PFA fixed and permeabilized in 0.2% Triton X-100 prior to blocking in 3% BSA for 1 hour at 37°C. The primary antibody was diluted 1/200 and incubated with the sample for 12 hours at 4°C. An Alexa Fluor® 488 conjugated goat anti-rabbit antibody was used as the secondary.
