Question 1

Monday 26-March-2012

Dear Technical Team,

Hope this e-mail finds you well!

Kindly suggest the exact concentration of Rabbit polyclonal Secondary Antibody to Mouse IgG - H+L (HRP) (ab6728) since on it's datasheet the concentration is mentioned as 1 mg at 2mg/ml .

Looking forward towards your valued response on it.

With Best Regards,

ANSWER:

Thank you for your message.

I am pleased to help answer your question.

I can confirm that '1 mg at 2mg/ml' means that the vial contains 1 mg of antibody at a concentration of 2 mg.ml. The concentration is 2 mg/ml. (Therefore the vial contains 500 ul).

I hope this will be helpful to you. If you have any further questions, please do not hesitate to contact us.

Question 2

Friday 23-March-2012

I'm confused because when I had inquiry about the precipitation before, I received emailusing them with gentle vortex but,
this time, you answered spin down it and use only supernatant.
Let me know which method is right.

Best regards,

ANSWER:

Thank you for your reply.

I can confirm that for the HRP conjugated antibodies the approach that Irecommended is the correct one. Spining the vial down will prevent free HRP to bind to un-specifically to samples and therefore avoid increased background.

If the antibody is not purified and therefore also not conjugated then we would recommend gentle vortexing. This could be whole antiserum for example.
In this casethe antibody does contain proteins and other solutes it would not be abnormal to see a little cloudiness or precipitation, especially after being stored at a low temperature and/or for a long period of time. Most of the time bringing the antibody up to room temperature, mixing it well and then diluting it to a working concentration will help incorporate any protein aggregates and precipitation.

I hope this information will explain the different suggestions we gave and I apologise for the confusion.

Question 3

Thursday 22-March-2012

Dear technical team,

We usually recommendab6721, ab6741, ab6789 and ab6728 secondary antibodies because those have many references,
and many customers use them now.
But, there's some precipitation, sometimes we have complain about that.
So, I would like to know only those cat# haveprecipitation or other HRP antibodies also show precipitation.

Best regards,

ANSWER:

Thank you for your enquiry. I am sorry to hear the customer has concerns regarding the quality of ab6721, ab6741, ab6789 and ab6728.

As the antibody does contain proteins and other solutes it would not be abnormal to see a little cloudiness or precipitation, especially after being stored at a low temperature and/or for a long period of time.

If HRP precipitates I can suggest to spin down the vial and only use the supernatant. The quality of the antibody is usually not compromised. Precipitates of HRP can happen with all HRP conjugated antibodies and we are monitoring this closely to make sure the product quality does not suffer.

Please do not hesitate to contact us again ifan antibody does not work.

Thank you for your time and I hope I could reassure you that ab6721, ab6741, ab6789 and ab6728 are of good quality.

Question 4

Tuesday 24-January-2012

I am interested in purchasing a secondary antibody for use in immunohistochemistry that can work with the following primary antibodies from your company (ab24590, ab7817, ab955, and ab89064). I have searched the catalog and found ab6728 and ab6788 as possible candidates for this purpose. Could you kindly suggest on which one would work better with those primary antibodies for immunohistochemistry (IHC-P)?

Thank you,

ANSWER:

Thank you for contacting Abcam.

I feel that both products are suitable for IHC-P. Both have many specific references and in-depth scientific support data right on the datasheet. They differ mainly in the species in which they were raised and in the tags used for detection. Ab6728 is an HRP conjugate whereas Ab6788 is Biotin labeled. I believe that you should choose the secondary which has an associated detection system that you are most familiar with in case you should run into any staining issues.

I hope this information is helpful. Please let me know if you have any questions.

Question 5

Thursday 08-December-2011

Dear TechSupport, We distribute your products in Russia and one of customers complained on product ab6728 recently. He spotted and yellowing jelly-like precipitate and wondering what can it be and if it is safe using the product. The picture is enclosed. Order reference for that purchase is ######. I will look forward for your reply.   With kind regards,

ANSWER:

Thank you for your enquiry.

I am sorry to see the customer has some concerns regarding the quality of this antibody. I understand their reservations.

Reviewing our records, it is quite common for this antibody to contain some precipitate and we are continuing to monitor this. In our customers experience, it will usually still will work well in experiments. I can recommend for the customer to mix the antibody well with a pipette, and use in their experiments as usual. If they do encounter any difficulties with their results, then please do not hesitate to get back to me as soon as possible with a completed technical questionaire. This product will be covered by our Abpromise guarantee in the applications and species listed on the datasheet.
