Question 1

Friday 20-April-2012

I would like to try using the following antibody in a IP however the concentration of the antibody is not displayed on the data sheet provided or online.
Can you help?
Anti Radixin [AF1H1] AB85353
Thanks

ANSWER:

Thank you for contacting us.

I apologise for the delay, I contacted my lab colleagues to check whether this antibody has ever been tested in IP applications. They confirmed it was once used in the past with bad results, therefore we wouldn’t recommend it for this application.

However, there are two other antibodies against the same target already tested – and therefore guaranteed – in IP. Here are the links to their datasheets:

http://www.abcam.com/Radixin-antibody-EP1862Y-ab52495.html

http://www.abcam.com/Radixin-antibody-ab50238.html

I hope they fulfil your requirements.

Regarding ab85323’s concentration I am afraid it cannot be determined. This monoclonal antibody is produced by ammonium sulphate precipitation from ascites. Different lots of this unpurified antibody preparation vary significantly their protein concentration and only a fraction of the proteins is the specific antibody.

It can be assumed that the concentration of the total protein for ab85323 is 0.5-2 mg/ml considering the band size on QC test.

I hope this information helps, and I am very sorry for the delay. Please do not hesitate to contact us if you need further clarification.

Question 2

Inquiry: Antibody concentration?

ANSWER:

Thank you for contacting us.

Unfortunately, the exact concentration of the Anti-Radixin antibody [AF1H1] cannot be determined.

This monoclonal antibody isproduced by ammonium sulfate precipitation from ascites. Different lots of this unpurified antibody preparation vary significantly their protein concentration and only a fraction of the proteins is the specific antibody. It can be assumed that the concentration of the total protein for ab85323is 0.5-2 mg/ml considering the band size on QC test. However, the specific antibody concentration is unknown, and you may refer to our 'concentration estimates' (attached) as a guideline for estimation. In these cases we focus on theantibody’s activity for each application and recommended dilutions are stated on the datasheet. Please remember that these dilutions and concentration estimates are recommended as a starting point, and it may be necessary to adjust the dilution based on the experimental results.

I am sorry that I cannot be of more assistance on this occasion, but Ihope this information is nevertheless helpful to you. Please do not hesitate to contact us if you need any more advice or information.
