Our Abpromise guarantee covers the use of ab86267 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
WB: Use a concentration of 1 µg/mlDetects a band of approximately 66 kDa (predicted molecular weight: 66 kDa).
ICC/IF: Use a concentration of 5 µg/ml
Essential subunit of N-oligosaccharyl transferase enzyme which catalyzes the transfer of a high mannose oligosaccharide from a lipid-linked oligosaccharide donor to an asparagine residue within an Asn-X-Ser/Thr consensus motif in nascent polypeptide chains.
Expressed in all tissues tested.
Belongs to the OST1 family.
Endoplasmic reticulum membrane. Melanosome. Identified by mass spectrometry in melanosome fractions from stage I to stage IV.

Western blot - Ribophorin I antibody (ab86267)

Anti-Ribophorin I antibody (ab86267) at 1 µg/ml + Transfected 293T cell lysate at 10 µg
Secondary
anti-Rabbit IgG HRP at 1/50000 dilution
Predicted band size : 66 kDa
Observed band size : 66 kDa
Immunocytochemistry/ Immunofluorescence - Anti-Ribophorin I antibody (ab86267)

ICC/IF image of ab86267 stained HeLa cells. The cells were 4% formaldehyde (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab86267, 5µg/ml) overnight at +4°C. The secondary antibody (green) was ab96899 Dylight 488 goat anti-rabbit IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
