Dear Abcam, I had previously ordered S100A9 peptide (ab74434) and Anti-S100A9 antibody (ab50143). Dot blotting was done using these 2 products. the anti-S100A9 antibody did bind to my samples however it did not bind to the S100A9 peptide (ab74434) supplied by you. It says in the Anti S100A9 antibody (ab50143)description that the antibody with bind to the full length native protein however it did not bind to S100A9 peptide (ab74434). it would be great if you could look into this matter and let me know the possible reasons why the antibody did not bind to the S100A9 peptide(ab74434). Thank you -- |

Thank you for contacting us. The immunogen for ab50143 is full length S100A9 (113 amino acids). However, the epitope for the antibody is not mapped. Since it is a monoclonal antibody, it can only recognise a single epitope along the full length protein. ab74434 is a 17amino acid peptide derived from S100A9. The reason why ab50143 can recognise your sample but not the peptide (ab74434), it is probably because the peptide does not contain the epitope of the antibody. For ab74434, I would suggest to use it with another antibody ab63818. I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information. |
