Question 1

Friday 16-September-2011

Dear Thomas,

We are interested in the anti-SAFB antibody cat # ab8060 for a western blot in human cells.

Thank you very much for your help.

ANSWER:

Antibody ab8060 will be useful for staining blots of your human cell lysates. However, there may have been some mis-communication regarding when we are able to offer the testing discount that was offered to your colleague.

For tested applications and species (listed on the datasheet), our Abpromise guarantee applies: if the antibody fails, we will suggest modifications to the protocol if we feel they are necessary, and if we are unable to improve the result we will offer a replacement or credit or refund, if you contact us about the issue within 6 months of the purchase. For ab8060, western blotting of human samples is covered by our guarantee.

For untested applications or species (for instance, if you wanted to use ab8060 on zebrafish samples or in FACS), our guarantee does not apply, but we do offer the testing discount program: test the antibody on the untested application or species, send us your results in the form of an Abreview (www.abcam.com/abreviews), and we will in turn offer you a credit for a free antibody. Please note that you will need to be issued a discount code before placing you order.

If you have any questions, please contact me.

Question 2

Thursday 30-October-2003

I am currently using the SAF-B primary antibody (ab8060(6F7)). I have noticed that the amino acid sequence of the immunogen on the Product Datasheet (PEARDKEDGRKF) is slightly different to the NCBI entry for SAF-B where the corresponding protein sequence is PEARDSKEDGRK, where there is an additional S residue. Is this just a printing error? Also the Product Datasheet says the antibody cross-reacts with human and mouse and as I have not yet been able to recreate the mouse cross-reaction in Western blots, could you confirm this as also the same antibody from Upstate biotechnology (cat# 05-588) says it only cross-reacts with human and not mouse?

ANSWER:

Thank you for your enquiry. You were correct in pointing out the accidental omission of the serine residue from the listed immunogen sequence. I apologize for this error and I have updated the datasheet. The correct peptide sequence is as follows: PEARDSKEDGRKF (as was used for the generation of polyclonal antibodies in JCB 67:275, 1997).

We produce some of our antibodies in-house and others are sourced from suppliers whose products we believe will be of interest to our customers. This product falls into the latter category. The identity of our suppliers remains commercially sensitive information. However, I can tell you that the antibody was not obtained from Upstate.

If you have any more questions, please contact us again. Also, if you would like to provide us with more details regarding the problems you are experiencing with the Westerns, we can do our best to help you out.
