Antibody ab8060 will be useful for staining blots of your human cell lysates. However, there may have been some mis-communication regarding when we are able to offer the testing discount that was offered to your colleague.
For tested applications and species (listed on the datasheet), our Abpromise guarantee applies: if the antibody fails, we will suggest modifications to the protocol if we feel they are necessary, and if we are unable to improve the result we will offer a replacement or credit or refund, if you contact us about the issue within 6 months of the purchase. I am currently using the SAF-B primary antibody (ab8060(6F7)). I have noticed that the amino acid sequence of the immunogen on the Product Datasheet (PEARDKEDGRKF) is slightly different to the NCBI entry for SAF-B where the corresponding protein sequence is PEARDSKEDGRK, where there is an additional S residue. Is this just a printing error? Also the Product Datasheet says the antibody cross-reacts with human and mouse and as I have not yet been able to recreate the mouse cross-reaction in Western blots, could you confirm this as also the same antibody from Upstate biotechnology (cat# 05-588) says it only cross-reacts with human and not mouse?

Thank you for your enquiry. You were correct in pointing out the accidental omission of the serine residue from the listed immunogen sequence. I apologize for this error and I have updated the datasheet. The correct peptide sequence is as follows: PEARDSKEDGRKF (as was used for the generation of polyclonal antibodies in JCB 67:275, 1997). We produce some of our antibodies in-house and others are sourced from suppliers whose products we believe will be of interest to our customers. This product falls into the latter category. The identity of our suppliers remains commercially sensitive information. However, I can tell you that the antibody was not obtained from Upstate. ab8060 at 1/500 staining human U2OS cells (osteosarcoma cell line) by ICC/IF. The cells were paraformaldehyde fixed and blocked with BSA prior to incubation with the antibody for 30 minutes. A FITC conjugated goat anti-mouse antibody was used as the secondary (right hand panel). The cells were also stained with DAPI (left hand panel).
