Relevance
|
SAFB (Scaffold Attachment Factor-B) is a member of a subset of nuclear matrix proteins that mediate the attachment of chromatin to nuclear protein complexes however it is not part of the nuclear matrix.
SAFB is ubiquitously expressed and appears to be a multifunctional protein involved in chromatin organization, mRNA processing and transcriptional regulation. It is expressed at high levels in the CNS and at low levels in the liver. Also expressed in a wide number of breast cancer cell lines.
SAFB has been demonstrated to interact with and function as a corepressor with many nuclear receptors such as estrogen receptor-alpha, PPAR-gamma, and FXR-alpha.

SAFB antibody [6F7] (ab8060) at 1/5000 dilution + 293T cells - whole cell lysate at 30 µg
Secondary HRP conjugated Sheep anti-mouse IgG developed using the ECL technique
Performed under reducing conditions.
Observed band size : 130 kDa (why is the actual band size different from the predicted?) Additional bands at : 60 kDa (possible non-specific binding).

Immunocytochemistry/ Immunofluorescence - SAFB antibody [6F7] (ab8060)
ab8060 at 1/500 staining human U2OS cells (osteosarcoma cell line) by ICC/IF. The cells were paraformaldehyde fixed and blocked with BSA prior to incubation with the antibody for 30 minutes. A FITC conjugated goat anti-mouse antibody was used as the secondary (right hand panel). The cells were also stained with DAPI (left hand panel).
