Recommended dilutions
IP: 1 µg immunoprecipitates SAP30 from 500µg of yeast extract.
WB: Use at a concentration of 1 - 4 µg/ml. Detects SAP30 in a yeast extract, proteins can be visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. Predicted molecular weight: 27 kDa, however ab15035 crossreacts with numerous additional bands greater than 50kDa in a yeast extract.

Not tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.

Relevance 

Hypoacetylation of core histones is believed to be one mechanism of achieving repression of gene transcription. In human cells, a large multiprotein complex containing the transcriptional repressor Sin3, multiple histone deacetylases (HDAC1 andHDAC2), histone binding proteins (RbAp46 and RbAp48), and other polypeptides, such as SAP30 mediates histone de-acetylation. SAP30 interacts directly with multiple components of this complex, most notably with HDAC1. SAP30 may regulate histone deacetylase activity and substrate specificity. Not all HDAC1-containing complexes contain SAP30 and in yeast Sap30 can recruit the deacetylase complex to promoter DNA in a Sin3 independent manner. Furthermore, SAP30 immunoprecipitates contain reduced deacetylase activity towards histone H3 relative to HDAC1 immunoprecipitates.
