ab51502 staining cultured rat cortical neuron cells by ICC/IF.  Cells were PFA fixed, permeabilized in 0.1% triton and blocked with 10% BSA for 1 hour at 22°C.  The sample was then incubated with ab51502, diluted 1/25 for 2 hours at 22°C prior to incubation with a Cy3® conjugated donkey anti-mouse antibody, diluted 1/400.  In the image, red is SATB2 and green is F-actin

Immunohistochemistry (Frozen sections) - SATB2 antibody [SATBA4B10] (ab51502)

ab51502 staining SATB2 in mouse brain tissue sections by IHC-Fr (Frozen sections). Tissue samples were fixed with paraformaldehyde and blocked with 10% serum for 1 hour at 22ºC. The sample was incubated with primary antibody (1/500) at 4ºC for 16 hours. An Alexa Fluor^®488-conjugated Goat polyclonal to mouse IgG (1/1000) was used as secondary antibody.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - SATB2 antibody [SATBA4B10] (ab51502)

ab51502 staining SATB2 in human fetal cortex tissue by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections). Tissue was fixed with paraformaldehyde and a heat mediated antigen retrieval step was performed using 10mM citrate buffer pH 6.0. Samples were then blocked with 10% serum for 1 hour at 25ºC followed by incubation with the primary antibody at a 1/100 dilution for 24 hours at 4ºC. A Cy3®-conjugated horse anti-mouse polyclonal was used as secondary antibody at a 1/200 dilution. Counter-stained with DAPI (blue).

Immunocytochemistry/ Immunofluorescence - SATB2 antibody [SATBA4B10] (ab51502)

ab51502 staining SATB2 (red) in a mouse cortex e17.5 cells by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with paraformaldehyde, permeabilized with Triton X-100 and blocked with 3% Donkey serum for 20 minutes at 22ºC. Samples were incubated with primary antibody (1/10) for 90 minutes at 22ºC. An Alexa Fluor^®555-conjugated Donkey anti-mouse polyclonal (1/200) was used as the secondary antibody.
Green: CTIP2 (ab18465) staining
Blue: TBR1 (ab31940) staining
