Target
- FunctionThis magnesium-dependent enzyme catalyzes the hydrolysis of ATP coupled with the translocation of calcium from the cytosol to the sarcoplasmic reticulum lumen. Contributes to calcium sequestration involved in muscular excitation/contraction.
- Tissue specificitySkeletal muscle, fast twitch muscle (type II) fibers.
- Involvement in diseaseBrody myopathy (BRM) [MIM:601003]: A disorder of muscle function that is characterized by painless muscle cramping and exercise-induced impairment of muscle relaxation. Note=The disease is caused by mutations affecting the gene represented in this entry.
- Sequence similaritiesBelongs to the cation transport ATPase (P-type) (TC 3.A.3) family. Type IIA subfamily.
- Developmental stageIsoform SERCA1A accounts for more than 99% of SERCA1 isoforms expressed in adult skeletal muscle, while isoform SERCA1B predominates in neo-natal skeletal muscle.
- Cellular localizationEndoplasmic reticulum membrane. Sarcoplasmic reticulum membrane.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-SERCA1 ATPase antibody (ab124501)IHC image of ab124501 staining in human skeletal muscle formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab124501, 1µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.
