Primary antibody notes

SIPA1 is a mitogen induced GTPase activating protein (GAP). It exhibits a specific GAP activity for Ras-related regulatory proteins Rap1 and Rap2, but not for Ran or other small GTPases. This protein may also hamper mitogen-induced cell cycle progression when abnormally or prematurely expressed. It is localized to the perinuclear region.

GTPase activator for the nuclear Ras-related regulatory proteins Rap1 and Rap2 in vitro, converting it to the putatively inactive GDP-bound state.

Tissue specificity

Expressed in fetal as well as in adult tissues. Expressed abundantly in the lymphoid tissues such as thymus, spleen and peripheral blood lymphocytes and also shows a significant expression in the spinal cord.

Predicted band size : 130 kDa


20µg of B cell lysate from different patients with Chronic Lymphocytic Leukaemia (a B cell malignancy) were separated by SDS-PAGE and blotted with ab10162 for 1 hour (1/500 dilution). The levels of SIPA1 in patients with CLL was compared to that in normal B cells (end lane).

Spa-1 is the Rap1-GAP of lympocytes, and therefore all the samples should all possess Spa-1.  Equal protein was loaded in each lane. A band of 130kDa was detected in all samples (arrow). Non-specific bands of ~ 60kDa were also detected.
