Our customer would like to purchase ab123457 and has inquiry the meaning of "recovery" in the protocol booklet p19 and datasheet..

"serum" mentionedon recovery table, the customer believes serum sample can be applied this product.

The reference to “recovery” in the protocol book of SIRT1 Human ELISA Kit (ab123457) might also be called “interference”. The point of this experiment is to validate the ELISA kit to ascertain how certain substances when added to the samples effect the finalELISA signal. In this specific example, goat serum was shown to not have a significant effect on the ELISA assay outcomein measuring recombinant SIRT1 (the values of recombinant SIRT1 measured in the presence of serum were 98-112% the values of recombinant SIRT1 measured without serum added).This means that the assay should be suitable for measuring SIRT1 in serum samples. Please note however that thiskit is for use in measuring human SIRT1 only.


Whilst the kit has not been tested using human serum samples, we cannot see a reason why it should not work. If using serum samples with the kit, we would suggest diluting the serum in extraction buffer at various ratios until the recovered signal is within the detection range of the standard curve. We would suggest initially not exceeding 10 % serum, but if the customer gets no signal at this level they could try higher levels.

Depending on how confident the customer is in detecting SIRT1 in human serum, they could also first try western blotting with the serum sample using theanti-SIRT1 antibody(ab110304) which is provided with the kit but can also be purchased separately. However this will be less sensitive than the ELISA assay.
