I was wondering if you could possibly help me. I have used four of your

antibodies for IHC:

SIRT1 (ab32441)

SIRT4 (ab10140)

SIRT5 (ab49173)

SIRT7 (ab78977)

In order to ensure the specificity of these antibodies before publishing

any results I was hoping to perform blocking peptide experiments for

each of the antibodies. I noticed that you have a suitable blocking

peptide for SIRT4 (cat# ab23185). However, I have been unable to find to

suitable blocking peptides for the SIRT1, 5 and 7 antibodies that I have

used. Do you have a suitable blocking peptide for each of these

antibodies? If these are unavailable would it be possible to get the

necessary peptides made?

Thank you for contacting us with your question. We do have the blocking peptide for the SIRT5 antibody ab49173, but it isn't on our website at this point so I'm working to have this added. I'll have to check with the lab about the peptide for the SIRT7 antibody ab78977, and I'll get back to you once I have more information about its availability. Unfortunately, the peptide for the SIRT1 antibody ab32441 is not available for sale. I will be in touch once I hear about the ab49173 and ab78977 peptides, but please let me know if you have any questions in the meantime.

Phone call: customer kindly asked for anti SIRT1, SIRT2 and SIRT6 antibodies that can used in immunohistochemistry.

SIRT6 The human, mouse and rat proteins are homologous in N terminal and central region of protein. So the antibodies available to these regions will most likely cross-react. ab62739, cross reacts with human, mouse and rat. Not yet tested in Immunohistochemistry. 

ab105391, cross reacts with human, mouse and rat. Not yet tested in Immunohistochemistry. These antibodies are not yet tested in Immunohistochemistry. However this does not mean that these are not suitable for IHC it only mean we do not have tested data to confirm their use. If you are interested in testing one of these in IHC then let me know I will send you the Abreview discount code;  which states we will offer a next free antibody in exchange of an antibody review.   

SIRT1 ab32441 and ab32424 These antibodies are tested in IHC so are fully guaranteed to work in IHC. The mouse and rat species is predicted which is based on sequence homology of immunogen between these species. If you are interested in using these antibodies in mouse and rat then let me know I will provide you the Abreview discount code which will give you your next purchase of an antibody free of charge. ab75435 and ab110304 does cross react with human, mouse and rat however not tested in IHC therefore these are eligible for Abreview discount programme.

I have tried the new step of 10 minute permeabilisation step using 0.2% Triton/PBS), but i got the same result again i.e. cytoplasmic,membranous and nuclear staining. i am wondering if you could help me further with this. i would like to mention that i ran out of the antibody so i would appreciate a replacement as you kindly offered in your email
