Thanks for your reply. There are several serines between amino acid 25-55 of human sirt1. To my knowledge, at least it has been shown that serine 47 can be phosphorylated. Do you have an idea whether this antibody can recognize both unphosphorylated and phosphorylated forms of sirt1?

Hi, What is the concentration of the SIRT1 antibody (Cat. ab32424, lot: GR56401-2)? I was trying to use this antibody for intracellular staining, and got high background. Since you have intracellular staining data using this antibody on your website, I was just wondering if you can give me a detailed protocol. Thanks a lot and have a nice day!

Thank you for contacting Abcam. The concentration for ab32424, lot#GR56401-2 is 0****** mg/mL. I am sorry about the problems that you are having with this antibody in staining, below is the only protocol we have for using ab32424 in IF: ICC/IF image of ab32424 stained HepG2 cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab32424, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. If the above information does not prove helpful, please could you send me a copy of the protocol that you are using and I would be happy to see if I can make any suggestions. Please let me know if there is anything else I can help you with.

SIRT6 The human, mouse and rat proteins are homologous in N terminal and central region of protein. So the antibodies available to these regions will most likely cross-react. ab62739, cross reacts with human, mouse and rat. Not yet tested in Immunohistochemistry. 

ab105391, cross reacts with human, mouse and rat. Not yet tested in Immunohistochemistry. These antibodies are not yet tested in Immunohistochemistry. However this does not mean that these are not suitable for IHC it only mean we do not have tested data to confirm their use. SIRT1 ab32441 and ab32424 These antibodies are tested in IHC so are fully guaranteed to work in IHC.
