Recommended dilutions ChIP: Use at a concentration of 4 µg/ml. IHC-P: Use at an assay dependent dilution (PMID 19389353). ICC/IF: Use at a concentration of 3 µg/ml. IHC-Fr: Use at an assay dependent dilution. WB: Use at a concentration of 0.3 µg/ml. Detects a band of approximately 37 kDa (predicted molecular weight: 34 kDa).
In-house IHC-P testing of this antibody gave inconclusive results. However, we have received several positive reviews from customers using it in that application.
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

SOX 2 is also known as SRY related HMG BOX gene 2. All SOX proteins have a single HMG box and bind linear DNA in a sequence-specific manner, resulting in the bending of DNA through large angles. Bending causes the DNA helix to open for some distance, which may affect binding and interactions of other transcription factors. SOX1, SOX2 and SOX3 show the closest homology to SRY. They share maximum homology within the HMG domain and are expressed mainly in the developing nervous system of the mouse. These genes share significant homology outside the HMG box also and are highly conserved throughout their evolution.

Immunocytochemistry/ Immunofluorescence - SOX2 antibody - Embryonic Stem Cell Marker (ab15830)
ICC/IF image of ab15830 stained mouse embryonic stem cells. The cells were 4% formalin fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab15830, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue).

Immunofluorescence - SOX2 antibody - Embryonic Stem Cell Marker (ab15830)
Sox2-specific antibody, ab15830, stained the nucleus of human ES cells (H9 line). Upon differentiation into neurones ab15830, but not Oct4-specific antibody, stained the nuclei (data not shown). Mouse embryonic fibroblasts and ES cells differentiated into early mesoderm were not stained by ab15830. Dilution used: 5µg/ml. Ludovic Vallier, University of Cambridge
