Target
- FunctionTranscription factor that forms a trimeric complex with OCT4 on DNA and controls the expression of a number of genes involved in embryonic development such as YES1, FGF4, UTF1 and ZFP206 (By similarity). Critical for early embryogenesis and for embryonic stem cell pluripotency.
- Involvement in diseaseDefects in SOX2 are the cause of microphthalmia syndromic type 3 (MCOPS3) [MIM:206900]. Microphthalmia is a clinically heterogeneous disorder of eye formation, ranging from small size of a single eye to complete bilateral absence of ocular tissues (anophthalmia). In many cases, microphthalmia/anophthalmia occurs in association with syndromes that include non-ocular abnormalities. MCOPS3 is characterized by the rare association of malformations including uni- or bilateral anophthalmia or microphthalmia, and esophageal atresia with trachoesophageal fistula.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - SOX2 antibody (ab97959)Carl Hobbs, King`s College London, United KingdomIHC-P image of SOX2 (ab97959) on zebrafish developing retina sections. The sections were fixed in formaldehyde and underwent heat mediated Anigen retrieval using citric acis (pH6). The sections were then blocked in 1% BSA solution for 10 mins at 21°C. The upper section used Abcam's EXPOSE HRP-DAB polyvalent kit (ab80436). The lower section was immunostained using my std stABCpx-DAB method. -
ICC/IF image of ab97959 stained mouse embryonic stem cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab97959, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

Tissue was fixed with paraformaldehyde and antigen retrieval was by heat mediation in citrate buffer. Samples were incubated with primary antibody (1/500 in TBS + 1% BSA) for 12 hours at 4°C. An HRP-conjugated goat anti-rabbit polyclonal IgG (1/200) was used as the secondary antibody. -
