Question 1

Wednesday 04-April-2012

Do you have any papers to support that sry protein is present
in adult mouse testis. I thought it was embryonic testis?

ANSWER:

Thank you for your reply.

I apologize for any confusion. We do not carry embryonic mouse testis lysates. I can confirm that the WB image is from fetal mouse, so I will update the image caption to avoid further confusion.

According to Swiss Prot:

Expressed in the substantia nigra of the brain (at protein level). Expressed in diencephalon, cortex, the substantia nigra of the midbrain and the medial mammillary bodies of the hypothalamus of male, but not female.

Expressed in gonadal somatic pre-Sertoli cells from 10.5 to 11.5 dpc. Expressed in pre-Sertoli cells located centrally in the genital ridge and then later in cells located at the cranial and caudal poles (at protein level).

Here is one article about adult testis expression of sry, but unfortunately I do not have access to the full text and do not know enough about the protein to comment definitively about expression during different development stages.

I hope this information helps. Please contact us with any other questions.

Question 2

Tuesday 03-April-2012

Thanks for your response. I did order this
antibody. I was wondering if you can send
me a positive control (testis sample etc.) which I can use
to validate if this antibody actually works. I may not see
the signal in the tissue of my interest and will never know
without a proper positive control if the antibody did not work/
it is just a problem of no expression in my sample.
Thanks for you support!

ANSWER:

Thank you for your inquiry.

We do sell the positive control mouse testis whole cell lysate as ab29289, but this is something that you would have to purchase.

Question 3

Thursday 29-March-2012

I am kind of confused. Sry is a pretty big protein, it would be good to know
if this antibody is raised against a peptide mapping to which amino acids
(for example 105-129 etc.) of mouse SRY.
I am asking this as there has been a lot of concern regarding mouse SRY
antibody and there are scientific groups making their own antibody as
the commercially available antibodies are non specific.

ANSWER:

Thank you for your reply.

Mice were immunized with a full length SRY protein and then monoclonal antibodies were derived from that. It is not uncommon to not map the specific sequence of amino acids that is targeted by the monoclonal antibody.

This antibody is guaranteed by our Abpromise and we will happily replace or refund any product that does not perform as stated on the datasheet when reported within 6 months of purchase.

I hope this information is helpful. Please do not hesitate to contact us if you have any additional questions or concerns.

Question 4

Wednesday 28-March-2012

This is in regards to this SRY antibody. What peptide sequence was used as
an immunogen for this antibody.
Thank you.

ANSWER:

Thank you for contacting Abcam regarding ab22166.

The immunogen is the full length protein fused with a proprietary tag. The specific epitope has not been mapped so we do not know where the antibody specifically binds to the protein.

I hope this information is helpful. Please do not hesitate to contact us if you have any additional questions.

Question 5

Thursday 17-November-2011

I have a question about your antibody, code nr ab22166, can I use it for cytofluorimetry experiments? Can I consider SRY a male antigen? I have to check donor male cells in a female recipient. If no, have you an antibody to use for my purpose? Thank you

ANSWER:

Thank you very much for your interest in ab22166.

This should be a male sex-specific protein. http://www.uniprot.org/uniprot/Q05738
