Recommended dilutions Peptide ELISA: Antibody detection limit dilution 1/2,000. WB: Use at a concentration of 1 - 3 µg/ml. Detects a band of approximately 38 kDa (predicted molecular weight: 24 kDa). The observed molecular weight corresponds to earlier findings in the literature (Taketo et al, Dev Dyn. 2005 Jun; 233(2):612-22.; PMID: 15789443).
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Relevance

The Sex Determining Region (SRY) is a gene that encodes a transcription factor that is a member of the high mobility group HMG box family of DNA binding proteins. The gene is located on the short arm of the Y chromosome and has a gene map locus - Yp11.3. Common synonyms of the gene name are Testis Determining Factor (TDF) and Testis Determining Factor on Y (TDY). The main function of the SRY protein is to initiate male sex determination as a transcriptional activator, which regulates a genetic switch in male development. The SRY HMG box recognizes DNA by partial intercalation in the minor groove. The SRY protein then binds to the DNA and distorts its shape (SRY protein in action). In doing so, it regulates genes that control the development of the testes. Transcriptional activator which regulates a genetic switch in male development. It is responsible for initiating male sex determination. SRY HMG box recognizes DNA by partial intercalation in the minor groove. Defects in SRY are a cause of gonadal dysgenesis XY female type (GDXY); also known as 'XY females' or Swyer syndrome. Patients are found to have a 46,XY karyotype. They suffer rapid and early degeneration of their gonads, which are present in the adult as 'streak gonads', consisting mainly of fibrous tissue and variable amounts of ovarian stroma. As a result these patients do not develop secondary sexual characteristics at puberty. The external genitalia in these subjects are completely female, and Muellerian structures are normal. In contrast, subjects with 46,XY partial gonadal dysgenesis have ambiguous genitalia, a mix of Muellerian and Wolffian structures, and dysgenic gonads. Defects in SRY may be a cause of Turner syndrome; also known as Ullrich-Turner syndrome. Turner syndrome has its basis in a 45,X chromosomal aberration. It is characterized by gonadal dysgenesis with short stature, 'streak gonads', variable abnormalities such as webbing of the neck, cubitus valgus, cardiac defects, low posterior hair line. The phenotype is female. Contains 1 HMG box DNA binding domain.
