Thank you for the email. We have used the attached protocol with abcam SSEA4 in the past with good results. Please see attached images of past successful staining using the same protocol and SSEA4 product number. Also, please note the abcam Oct4 worked well using the same protocol this time and in the past. We have also performed a serial dilution, recently to try dial in the right concentration on the current lot with no luck.

Can you recommend a better SSEA4 lot? Have others reported any issues with this lot? Is there anything we are overlooking with our attached protocol that does not work for the recent SSEA4 lot where it has worked with others in the past?

I have reviewed your protocol and nothing stands out as being an issue. Your protocol appears to be standard and takes all the appropriate factors into account in terms of fixation, permeabilization, blocking, etc. In looking through our records, we have not received any other reports of this product not working for other users.

I am contacting you regarding Abcam SSEA4 MC813 ab16287. We are getting nonspecific staining with the SSEA4 stain. Please see attached image, green stain (both feeders and hES colonies are stained). Have you had any issues with this particular lot? We have performed a serial dilution and additionally had 12 researchers verify our results of nonspecific staining.

I am trying to isolate mouse MSC from fetal mouse liver (day 14 of pregnancy) using their ability to adhere to the plastic surface. I red that mouse MSC are positive for stage-specific embryonic antigen (SSEA-4). Also that anti-human SSEA-4 react with the mouse antigen. Do you know if it is possible to isolate mouse MSC using MACS with SSEA-4 positive selection? Can I use this antibodies like a marker of MSC for FACS?

I want to know the prise for unlabelled antibodies, of if you have them with magnetic beeds, and the antibodies for FACS (for Chanel 3, becouse my cells are eGFP +)

Abcam antibody ab16287, clone MC813, can be used for FACS with mouse cells and will recognize mouse SSEA-4. The literature reports that SSEA-4 is only expressed on mouse ES cells upon differentiation, and is not detected on undifferentiated mouse ES cells.
