Please see attached images of past successful staining using the same protocol and SSEA4 product number. Also, please note the abcam Oct4 worked well using the same protocol this time and in the past. We have also performed a serial dilution, recently to try dial in the right concentration on the current lot with no luck. |

I am trying to isolate mouse MSC from fetal mouse liver (day 14 of pregnancy) using their ability to adhere to the plastic surface. I red that mouse MSC are positive for stage-specific embryonic antigen (SSEA-4). Also that anti-human SSEA-4 react with the mouse antigen. Do you know if it is possible to isolate mouse MSC using MACS with SSEA-4 positive selection? Can I use this antibodies like a marker of MSC for FACS? I want to know the prise for unlabelled antibodies, of if you have them with magnetic beeds, and the antibodies for FACS (for Chanel 3, becouse my cells are eGFP +) |

Thank you for your enquiry. Abcam antibody ab16287, clone MC813, can be used for FACS with mouse cells and will recognize mouse SSEA-4. The literature reports that SSEA-4 is only expressed on mouse ES cells upon differentiation, and is not detected on undifferentiated mouse ES cells. Please consult the following reference for more information: Kannagi R et al. Stage-specific embryonic antigens (SSEA-3 and -4) are epitopes of a unique globo-series ganglioside isolated from human teratocarcinoma cells. EMBO J 2:2355-61 (1983). PubMed: 6141938 We have the unlabelled antibody for SSEA-4, catalogue number ab16287, but not a fluorochrome-conjugated or magnetic bead-conjugated version. 2102ep Human Embryonic Carcinoma cells were stained with SSEA4 antibody ab16287. As expected, staining localised to the cell surface (green). Nuclei are stained blue using Hoechst.
anti-SSEA4 antibody, ab16297 can be used as a marker of Embryonic Carcinoma cells in Flow Cytometry/FACS. As can be seen from the histograms, in non-differentiating conditions (i.e. without retinoic acid) NTERA2 cells were recognised by ab16297. However, upon differentiation (addition of retinoic acid), the antibody lost the ability to recognise the cells.
ab16287 staining SSEA4 in human Embryonic Stem Cells, HUES7 by Immunocytochemistry/ Immunofluorescence. Cells were fixed with paraformaldehyde, permeabilized with Triton and blocking with 10% serum for 1 hour was performed. Samples were incubated with primary antibody (1/100: in 1% serum, 0.1% Triton in PBS) for 1 hour at 37°C. An Alexa Fluor®588-conjugated goat polyclonal to mouse IgG was used at dilution at 1/100 as secondary antibody.
