STATs are Signal Transducers and Activators of Transcription
STATs are a family of cytoplasmic proteins that participate in cellular responses to cytokines and growth factors. Many cytokines involved in immune responses utilize the Jak-STAT signaling pathway. Jaks are receptor-associated protein tyrosine kinases, and STATs are activated by tyrosine phosphorylation. Abnormal signaling of the JAK-STAT pathway has been implicated in hematopoietic disorders including SCID and leukemia, and programming gene expression in biological events such as embryonic development, apoptosis, organogenesis, innate immunity, adaptive immunity and cell growth regulation. STAT1 knockout mice are defective in interferon-mediated functions.

Carries out a dual function: signal transduction and activation of transcription. Binds to the GAS element and activates PRL-induced transcription.

Post-translational
modifications
Tyrosine phosphorylated in response to IL-2, IL-3, IL-7, IL-15, GM-CSF, growth hormone, prolactin, erythropoietin and thrombopoietin. Tyrosine phosphorylation is required for DNA-binding activity and dimerization. Serine phosphorylation is also required for maximal transcriptional activity.

Anti-STAT5 antibody - ChIP Grade images:
ChIP - STAT5 antibody - ChIP Grade (ab7969)
ab7969 staining mouse skin epidermis whole tissue lysate by X-ChIP. The cross linking step was 10 seconds with 1% formaldehyde. The primary antibody was diluted 1/180 and incubated for 12 hours at 4°C.
ChIP dilution buffer: 16.7mm tris-cl (pH- 8.1) 0.01% SDS 1.1% triton-X-100 1.2mm EDTA, 16.7mm Nacl +1X complete protease inhibtor cocktail.
In the image, wild type (control) and TPA treated samples were immmunoprecipitated with rabbit IgG, pan RAR antibody or STAT5 antibody and the purified DNA was amplified using primers flanking the STAT5 binding element. Rabbit IgG and 2 other unrelated antibodies did not give any amplification.
PCR condition: 94, 30' 59, 30' 72, 45' 32 cycles. product size:310bp.
