Can you please send me the protocols used to test these antibodies in Flow Cytometry? Will trypsin present from releasing the cells from the culture plate interfere with the fluorophores? |
|||||||||
ANSWER: |
Thank you for your call earlier this week, and for your patience while I have been in touch with the lab. For both of these antibodies, you can follow the direct flow cytometry protocol on our website- http://www.abcam.com/index.html?pageconfig=resource&rid=11380 Though the optimal antibody concentration should be determined for your samples, for ab25323 we would recommend a starting concentration of 0.2 ug antibody per 10^6 cells. For 93567, we would recommend using between 0.25 and 0.5 ug of antibody per 10^5-10^8 cells. We also suggest removing the trypsin from the cell solution prior to staining. The cells can be re-suspended in ice-cold PBS with 10% FCS and 1% azide to ensure stability of the epitopes. I hope this information is helpful, but please let me know if there is anything else that we can do for you. |
