I found conflicting information about the expression of CD90 on NIH3T3 cells: No expression or expression of Thy1.2 (CD90.2). ab226 only recognizes Thy1.1 (CD90.1). I therefore suggest to use ab25672 (Anti-CD90 / Thy1 antibody [G7] (FITC)) which will bind CD90.1 and CD90.2. I could not find any reliable data on the expression of Ly6 on NIH3T3 cells either, but can confirm that ab25031 will bind to Ly6A.2 and Ly6E. I am sorry that I could not be more helpful, but I hope that the available literature in this area can provide some clarification. BALB/c spleen cells were activated by incubation for 48 hours with concanavalin A. After harvesting, the cells were stained with Rat anti-mouse Ly6A/E-FITC (less than or equal to 0.5 ug/106 cells) and activated cells were gated and analyzed on a FACScan flow cytometer.
ICC/IF image of ab25031 stained MEF cells. The cells were 4% PFA fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab25031, 5µg/ml) overnight at +4ºC. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
