Hello, I recently contacted abcam technical support about an antibody that did not work (see below). Would it be possible to apply the credit from this antibody toward the purchase of the following secondary antibody? Sheep polyclonal secondary antibody to mouse IgG (H&L), FITC, ab6804 Thank you, |

Thanks you for your email. Dr. Tanya is away this week so I am looking after her inbox.
I am sorry we are still waiting for questionnaire we have sent in last email. The refund or credit note can only be offered if a product was used in applications as stated on the datasheet and in case the scientific support team unable to resolve the issues.
This antibody is one of our best characterized antibodies which have been cited in many publications and Abreviews. We have not received any complaint about the batch you have mentioned so I am presuming that protocol optimization might be helpful in your experiment. So could you send us the protocol as soon as possible? If we see the protocol used is fine than we will either replace the product or will give you full refund.
Thank you very much for your cooperation in this case. We will look forward to hearing from you soon. |

My question is about anti-serotonin antibody. As you know, serotonin(5-HT)is a very small chemical which is supposed to freely diffuse within cells or organelles. In a standard immunostaining protocol, the sample tissue or cells should be permeabilized after fixation and washed for several times after antibody incubation. My concern is how is it possible that the serotonin remain its position and not be washed out of the cell? How does the anti-serotonin antibody work? It really bothers me. Thank you! |

ANSWER: |
Thank you for your enquiry. This antibody label cells expressing serotonin and not serotonin itself. In immunocytochemistry the antibody labels entero-chromaffine (EC) cells in colon. I have attached some publications on the subject should you require extra information. 1. ab16007 staining mouse intestinal tissue sections by IHC-Fr. The tissue was fixed with acetone, permeabilised by 0.1% TX-100 in AB-solution, and blocked with 10% serum for 30 minutes at 25°C. The primary antibody was diluted 1/200 in 10% normal goat serum in PBS, 0.1% TX100 and incubated for 24 hours at 4°C. A FITC-conjugated rabbit anti-mouse was used as the secondary.

Ab16007 staining of serotonin in human appendix by formalin-fixed and paraffin-embedded IHC. Note cytoplasmic staining of serotonin-secreting cells.
ab16007 staining Serotonin in pig small tissue section by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections). Tissue underwent paraformaldehyde fixation before heat mediated antigen retrieval in Citrate Buffer (pH 6) and then blocking with 10% serum was performed for 20 minutes at 22°C. The primary antibody was diluted 1/300 and incubated with sample in TBS/TX(0,3%)/BSA(1%) for 16 hours at 4°C. A Biotin conjugated goat polyclonal to mouse IgG was used at dilution at 1/200 as secondary antibody.
