Primary antibody notes

Ab16007 may be a useful tool for characterization of carcinoids. In a study of 48 cases of adenomas and carcinoid tumours of the middle ear, the antibody labelled tumour cells in 12 cases (25%). In 1 case of carcinoid tumour of the middle ear, the antibody labelled tumour cells. In a study of 43 cases of bronchial carcinoid tumours, the antibody labelled tumour cells in 13 cases (30%). Cells labelled by the antibody display a cytoplasmic staining pattern.

Relevance

Serotonin (5-hydroxytryptamine, or 5-HT) is a monoamine neurotransmitter synthesised in serotonergic neurons in the central nervous system and enterochromaffin cells in the gastrointestinal tract. Serotonin plays an important part in the biochemistry of depression, migraine, bipolar disorder and anxiety. It is also believed to be influential on sexuality and appetite. 5-HT is generally thought not to be released from synaptic terminal buttons in the manner of classical neurotransmission but from serotonergic varacosities into the extra neuronal space. From here it is free to diffuse over a relatively large region of space (>20µm) and activate 5-HT receptors located on the dendrites, cell bodies and presynaptic terminals of adjacent neurons. Serotonergic action is terminated primarily via uptake of 5-HT from the synapse. This is through the specific monoamine transporter for 5-HT, 5-HT reuptake transporter, on the presynaptic neuron. The pharmacology of 5-HT is extremely complex, with its actions being mediated by a large and diverse range of 5-HT receptors.

ab16007 mouse monoclonal [5HT-H209] used on cryopreserved ventral mesencephalic neurons dissected at embryonic day 14. Cells were thawed plated and grown for 14 days, fixed and stained with ab16007 at 1/50 overnight.

ab16007 staining mouse intestinal tissue sections by IHC-Fr.  The tissue was fixed with acetone, permeabilised by 0.1% TX-100 in AB-solution, and blocked with 10% serum for 30 minutes at 25°C.  The primary antibody was diluted 1/200 in 10% normal goat serum in PBS, 0.1% TX100 and incubated for 24 hours at 4°C.  A FITC-conjugated rabbit anti-mouse was used as the secondary.

ab16007 staining Serotonin in pig small tissue section by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections). Tissue underwent paraformaldehyde fixation before heat mediated antigen retrieval in Citrate Buffer (pH 6) and then blocking with 10% serum was performed for 20 minutes at 22°C. The primary antibody was diluted 1/300 and incubated with sample in TBS/TX(0,3%)/BSA(1%) for 16 hours at 4°C. A Biotin conjugated goat polyclonal to mouse IgG was used at dilution at 1/200 as secondary antibody.
