Synthetic peptide corresponding to the carboxy-terminus of the cloned rat Serotonin transporter.
ab79328 antiserum has been tested on tissue sections from the central nervous system. The staining pattern obtained with ab79328 corresponds to the pattern described using antiserum to serotonin itself. The serotonin transporter immunoreactivity corresponds to what has been described using other antibodies to the serotonin transporter. Preabsorption of ab79328 with the immunogen peptide completely abolishes the immunostaining.

Using a Cy3 conjugated secondary antibody. Fix tissue in a paraformaldehyde/picric acid-mixture (4% paraformaldehyde and 0.4% picric acid in 0.16 M sodium phosphate buffer, pH 6.9.
ICC/IF image of ab79328 stained PC12 cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody ab79328 at 1/100 dilution overnight at +4°C. The secondary antibody (green) was DyLight® 488 goat anti- guinea pig (ab96959) IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
