Dear Abcam, I was wondering if it is possible to order ab52903 in a smaller volume size (e.g. 20 ul) for evaluation purposes? I would like to trial this antibody for whole-mount immunofluorescence and 3-D multiphoton imaging in chick embryos. If the antibody is successful I would anticipate ordering in larger volumes. I would also be interested to know whether ab52903 has been tested for cross-reaction with other Smad family members, or if it is known/predicted to react with the chicken antigen. I am using the phospho-Smad3 antibody (ab52903) for immunocytochemistry experiments in smooth muscle cells. I have difficulty in finding a good fluorescent-labelled secondary antibody. I noticed that on your website you have two very nice images of immunocytochemistry/immunofluorescence. I was wondering if you could give me information about the secondary antibodies that were used in each of these two stainings. If not do you know of any other fluorescent-labelled secondary antibodies that can be used with Smad3 antibody for immunocytochemistry/immunofluorescence experiments? Preferably a secondary antibody that does not interfere with 488 wavelength.

Unfortunately I do not have the exact details of the secondary antibodies used for the ICC images on the front of the datasheet. I just searched for pre-adsorbed (recommended to reduce non-specific binding) secondary antibodies conjugated with a flurophore outside the 488 wavelength and identified 56 matches. I would encourage you to use the search engine on our website to identify suitable products, here is a link:

ab52903 staining Smad3 (phospho S423 + S425) in human TII Pneumocyte A549c ells by Immunocytochemistry/ Immunofluorescence. Cells were fixed with paraformaldehyde and permeabilized with 0.1% Triton x100 before blocking with 3% BSA for 1 hour at RT. Samples were incubated with primary antibody (1/200: in 3% BSA in 1x PBST) for 24 hours at 4°C. A TRITC-conjugated goat polyclonal to rabbit IgG was used as secondary antibody at 1/200 dilution.
