  • Application notesELISA: Use at a concentration of 0.5 - 1 µg/ml, this concentration is determined to detect recombinant human sTNF RII to a limit of 1.25 ng/well.
    Neut: Use at a concentration of 0.5 - 1 µg/ml, this concentration will block 50% of the bioactivity due to 0.3 µg/ml recombinant human sTNF RII, in the presence of 0.25 ng/ml TNF-alpha in a crystal violet cytolytic assay using mouse L929 cells.
    WB: Use at a concentration of 1 - 2 µg/ml, this concentration is determined using recombinant human sTNF RII at 50 ng/lane under non-reducing conditions.Predicted molecular weight: 75 kDa.

    Anti-Human sTNF RII is tested for its ability to neutralize the biological effect of sTNF-RII but not sTNF RI in a cytolytic assay using mouse L929 cells, in the presence of TNF-alpha. The ND50 of the antibody is defined as the concentration of antibody resulting in a one-half maximal inhibition of the soluble receptor activity on a responsive cell line, when the soluble receptor is present at a concentration just high enough to elicit a maximum response.

  • Target

    • FunctionReceptor with high affinity for TNFSF2/TNF-alpha and approximately 5-fold lower affinity for homotrimeric TNFSF1/lymphotoxin-alpha. The TRAF1/TRAF2 complex recruits the apoptotic suppressors BIRC2 and BIRC3 to TNFRSF1B/TNFR2. This receptor mediates most of the metabolic effects of TNF-alpha. Isoform 2 blocks TNF-alpha-induced apoptosis, which suggests that it regulates TNF-alpha function by antagonizing its biological activity.
