SorCS1 belongs to the family of VPS10 domain-containing receptors (the name is derived from the yeast vacuolar protein sorting protein-10, which is involved in sorting of carboxy-peptidase Y from the Golgi apparatus to the vacuole). SorCS1 immunoreactivity is widespread in a population of neurons throughout the brain. Two different types of cellular localization were observed. Most SorCS1 immunoreactive neurons exhibit a punctate cytoplasmic staining which extends into the dendrites, whilst occassionally SorCS1 neuronal immunoreactivity is associated with the plasma membrane.

Immunohistochemistry (PFA fixed) - SorCS1 antibody - Neuronal Marker (ab16641)

Immunofluorescent staining in rat hippocampus, using ab16641 SorCS1 antibody. [A] picture taken using a X10 objective and [B] X20 objective. Preincubation of antibody with the immunising peptide (ab16690) blocked the antibody from staining. ab16641 was used at 0.5µg/ml incubated overnight at room temperature. Secondary antibody used was anti-rabbit Alexa Fluor 488 at 1/1000 incubated for 2 hours at room temperature. Rat brain tissue was perfusion fixed with 4% PFA followed by overnight post-fixation in the same fixative, cryoprotected in 20% sucrose and frozen in OCT. 30µm coronal sections were cut on a cyrostat and immunohistochemistry performed by the 'free floating' technique.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - SorCS1 antibody - Neuronal Marker (ab16641)

IHCP-image of SorCS1 staining using ab16641(1:500) on mouse Hippocampus sections. The tissue was fixed in formaldehyde and the epitope on sections were revealed using heat mediated antigen retrieval using citric Acid (pH 6). the sections were blocked using 15 BSA for 10 mins.
