Thank you for contacting us. Indeed, neither ab68954 nor ab20920 have been tested for immunoprecipitation (IP) yet, but they could theoretically be used in this application. If it is just between the two, I would recommend ab20920 because it is an unconjugated antibody. The biotin-labeled antibody ab68954 might not bind to the Protein A or G agarose as well as the unconjugated antibody. Also, ab20920 seems to detect its antigen(s) in a rather native state (as it works with frozen tissue sections) which might be more suitable for IP. However, these antibodies are polyclonal antibodies against various soluble and structural antigens of the whole Staphylococcus aureus. By using the antibodies in IP, a variety of proteins would be pulled down. If this is not appropriate for your experimental setting, I would be pleased to assist you in finding a suitable antibody (for example against a certain target protein). If you decide to go ahead and purchase one of these products though, you might be eligible for our testing discount program. I have received a survey you have filled out relating your recent experience with ab20920. Reviewing my correspondance with you at the time I found that I had sent you a free replacement antibody to replace the faulty vial of ab20920 you received. I also offered to send you a complimentary free antibody. I am curious to know how the replacement vial I sent, ab68954, worked for you. In addition, you suggested waiting on the second free antibody until after you had tested ab68954. I would be happy to send you that second primary antibody at your convenience. I will be happy to help you resolve any remaining issues you man have with any of our products. |

sorry for my impricise question. But you are right. We don't need information about how the antibodies are tested, but how you test your clarified antibodies. That's because we have problems by producing lysate from S. aureus. We used different lysis methods and had no positive signals with your antibodies. Propably we don't use the right one? It would be kind if you let us know how you produce lysate from S. aureus to get the best signal with your antibody (a protocol of your method would be perfect).

Thank you for your enquiry. This antibody has been tested for use by immunofluorescence. It is yet to be determined whether it is suitable for western blot analysis. Therefore we have not generated a lysate for immunoblotting analysis and do not have a corresponding protocol that we can provide you with. I am sorry that I cannot be of more help on this ocassion. We do however have an antibody that HAS been shown to work by western blotting; Mouse monoclonal [332/423] to Methicillin Resistant Staphylococcus Aureus (ab35861). I am in touch with the source of this antibody to determine whether they can provide me with a protocol for its testing by western blotting. I appreciate your patience in this matter. |

The immunogen used to produce ab20920, Rabbit anti Staphylococcus aureus, is a mixture of intact organisms & soluble proteins from disrupted organisms. The cells were disrupted using a combination of sonication and freeze/thaw cycles. The immunogen preparation did not use any enzymatic methods. ab20920 staining an abscess in rat calf tissue prepared as a 20 µm cryoslice. Tissue was fixed in paraformaldehyde, permeabilised with 0,5% Trition X100 in PBS, and blocked with 10% serum plus 1% BSA for 1 hour at room temperature. ab20920 staining Staphylococcus aureus in human keratinocytes by Immunocytochemistry/ Immunofluorescence.Cells were fixed in methanol, permeabilized using 0.1% saponin/ PBS, blocked with 4% BSA for 30 minutes at 25°C and then incubated with ab20920 at 10µg/ml for 14 hours at 4°C.
