Question 1

Friday 13-April-2012

Do you have an extinction coefficient for product number ab63978, Streptolysin O protein?

ANSWER:

Thank you for contacting Abcam.
To find the extinction coefficient, I used the follwing link:
http://encorbio.com/protocols/Prot-MW-Abs.htm
For Streptolysin O protein ;
There are 571 Amino acids and Molecular Weight is 63.64 KiloDaltons
Charge is 3.5 assuming N and C termini are not blocked
There are 22 Tyrosines and 7 Tryptophans
The Molar Extinction Coefficient is 71280
The absorbance of a 1mg/ml solution at 280nm is 1.120
Please let me know if there is anything else I can help you with.

Question 2

Do you have any additional details about the protocol that is used to measure the activity of this protein?

ANSWER:

Thank you for contacting us.

The activity of this Streptolysin O proteinis measured by applying the protein to 3% sheep red blood cells in PBS, pH 7.4 at 37C for 30 minutes and counting the lysed cells under the microscope. Using this protocol,1HU is defined as the amount of protein required to produce 50% hemolysis.

I hope this helps, please let me know if you need any additional information.
