This antibody has been tested in Western blotting against equal loadings of purified wild type survivin, 2B and DEx3. When used at 1/500, it detects a single clean band of the appropriate size for 2B, but no band is detected with wild type survivin or DEx3.
We have not succeeded in detecting endogenous Survivin 2B using this antibody (Survivin 2B is expressed at very low levels).
This antibody has been used successfully however, to detect overexpressed Survivin 2B in whole cell extracts.

Recommended dilutions ICC/IF: Use at an assay dependent concentration. IF: 1/50. WB: 1/500. Found to work at 1/5000 dilution (see PMID: 16847456). Predicted molecular weight: 18 kDa. Can be blocked with Survivin 2B peptide (ab13762).
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

|
Survivin, a new inhibitor of apoptosis (IAP) protein, is expressed in the G2 / M phase of the cell cycle in a cycle regulated manner. At the beginning of mitosis, survivin associates with microtubules of the mitotic spindle in a specific and saturable reaction that is regulated by microtubule dynamics. Disruption of survivin microtubule interactions results in loss of survivin's anti-apoptosis function and increased caspase 3 activity, a mechanism involved in cell death, during mitosis. It indicates that survivin may counteract a default induction of apoptosis in G2 / M phase. The over-expression of survivin in cancer may overcome this apoptotic checkpoint and favour aberrant progression of transformed cells through mitosis. Survivin DEx3 (lacking exon 3) and survivin-2B (retaining a part of intron 2 as a cryptic exon) are isoforms of survivin that behave differently to survivin. Survivin-2B has acquired additional 23 amino acids encoded by the cryptic exon 2B. See Mahotka et al. 1999 for more details.Expressed only in fetal kidney and liver, and to lesser extent, lung and brain. Abundantly expressed in adenocarcinoma (lung, pancreas, colon, breast, and prostate) and in high-grade lymphomas.
