Question 1

Monday 06-February-2012

Hello sir/madam
recently i have tried the tap1(ab60112) in( Ihc-p) in oral squamous cell carcinoma.
and I want to question you about the antibody tap1(ab60112)
in the datasheet the cellular localization: cell membrane
why isthe cellular localization in cell membrane? The protein is localized in the membrane of endoplasmic reticulum in cell.
when i tried the primary antibody the colouris in the cytoplasma of theepithelial cells and sometime in the cell membrane.
is it normalthat the colour is inthe cytoplasma of the cells ?
thank you

ANSWER:

Thank you for your inquiry.

Indeed, I can confirm that TAP1 is a multi-pass membrane protein localised in the membrane of the endoplasmatic reticulum where it is said to form a pore, as stated on the datasheet and in SwissProt entry Q03518 (http://www.uniprot.org/uniprot/Q03518). Thus, the staining normally appears in the cytoplasm of the cells. Depending on factors such as the cell type, cell metabolic status and the section plane it sometimes may seem that the signal is in the cell membrane.

You may find examples of staining patterns for TAP1 in the Human Protein Atlas:
http://www.proteinatlas.org or more specifically http://www.proteinatlas.org/ENSG00000168394.
