Anti-TFIIB antibody [IIB8] (ab819)

I am writing you after so long that I am sure you will not remember my last e-mails dated from last November... I apologize for not replying soon after you sent me the antibody (order number 28372, TFIIB antibody (ab819)[IIB8]) but meanwhile it was not possible to go on testing the antibody. So, I am writing also to thank you for the batch (from lot 12091) you have sent to me, which has proven to be much more adequate to detect yeast TFIIB in my extracts (although not so efficient as the original lot 12091). Since, as you might remember, lot 25198 (TFIIB antibody (ab819)[IIB8]) was clearly unable to recognize TFIIB in my yeast extracts, and I still have some experiments to do using TFIIB antibody, I would be very grateful if you could inform me if it is still possible to order lot 12091 of that antibody.

We have looked at our database but did not receive any similar complaint so far. We are very sorry to inform you that we have sold out all the vials from Lot: 12091. However, we have new batch in stock which should work well.

Thank you for replying to my e-mail. I am afraid I need your help... I am worried about the fact this batch is no longer available, and I will explain you why:

In March 2003 I have order to Abcam the primary antibody TFIIB (ab819) [IIB8](lot 12091). The antibody was used in western blot to detect TFIIB in protein extracts prepared from Saccharomyces cerevisiae. The results show the expected correspondent band and therefore the antibody proved to be adequate for our studies. However, when a second vial was ordered in October, we have received a different lot (25198) that did not react with TFIIB under the same conditions previously tested even if the same protein extracts were used. Subsequently, I received a free-of-charge vial from lot 12091, and I recovered the band corresponding to TFIIB in my westerns.

Therefore, since I still have to do some urgent experiments using this antibody, I would like to know if you are aware of well-succeeded TFIIB batches against S. cerevisiae. I would really appreciate your help.

Thank you for your message. We have had no other complaints regarding this antibody or about other batches. I'm happy that you have had success with ab819 in the past and would like to reassure you that our policy is that if you happened to have problems with a future batch (using the antibody as stated on the datasheet) we will give you a replacement batch or refund.

However, we have not heard of this antibody being used on S. cerevisiae before. To our knowledge it has only been tested before on human and mouse, so we would suggest using human or mouse samples as a positive control. If you have any more questions, please contact us again.

Dear Sirs, We are currently using the ab819 antibody against TFIIB and we were able to sucessfully detect the yeast (S. cerevisiae) TFIIB.
