The majority of staining for TGF beta 1 will be in the extracellular space however you may see some staining of protein on its way out of the cell as its being processed.

Does the TGF-beta1 signals should locate in the cell membranes, plasma and nuclei in IHC assay?

The protein is secreted from the cell. It is then cleaved into a latency-associated peptide (LAP) and a mature TGFB1 peptide, and is found in either a latent form composed of a TGFB1 homodimer, a LAP homodimer, and a latent TGFB1-binding protein, or in an active form composed of a TGFB1 homodimer. The mature peptide may also form heterodimers with other TGFB family members.

I brought only one vial TGF beta1, so I don’t have the same antibody with another lot. However, I found the purchase order number. The abcam product was distributed by a Dutch company called ITK diagnostics BV in Uithoorn. The purchase order number of ITK diagnostics was AM-4243781.

Thanks for review. Yes, the 50 samples were all sections of the synovial sarcoma tissue.

Normally for nucleus staining, if they were not expected they might be chance that nonspecific.

Is there any way to explain why most of the samples, from the same tissue, using the same primary antibody and same protocol,stain as expected while some show nuclear staining?

We have inquiry to the attached antibody that we did not notice a consistent staining data as we expected from literature. We would like to understand perhaps some information, or advice from you?
