Question 1

Thursday 01-March-2012

I wish ask you about your antibody ab47068. I used it to find KIM-1 protein. I used lysate of A431 and Clear Renal Cells Carcinoma. I obtained just 2 bands in Clear Renal Cells Carcinoma: one around 29 kDa and a second one (much weaker than the first one) around 20 kDa. Your datasheet show a WB with an observed band size of 21 kDa (while the predicted band size is 39 kDa).
Could you help me to understand if my bands are aspecific or not?

ANSWER:

Thank you for contacting Abcam.

I believe that the band you are seeing at 21kDa is the correct size band that this antibody is supposed to recognize according to the datasheet. I am not sure what the 29kda band is, but if you are only seeing it in your Clear Renal Cells Carcinoma lysates, then it could be non-specific and you could clear this up by altering your blocking conditions. I have attached the Abcam western blot troubleshooting guide that may be of help to you.

Also I am not an expert on TIM1 but I did read somewhere that the full length protein (which would run at 39kDa) is further cleaved into a mature form and that could explain, why the predicted molecular weight is greater than what is actually seen. As I said though, I am not an expert on this protein and so I am sure that you know more about than I.

Please let me know if there is anything else I can help you with.
