Mouse monoclonal [3A4] to TIMP2
WB, IHC-Pmore details
Reacts with
Mouse, Rat, Rabbit, Guinea pig, Cow, Human
BALB/C mice were injected with a synthetic peptide from the N-terminal region of human TIMP 2.
This antibody gave a positive signal in Western Blot when tested against recombinant TIMP2 protein. In IHC, this antibody gave positive staining in a human pancreatic tumor section.

Our Abpromise guarantee covers the use of ab109708 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
WB: Use a concentration of 5 µg/ml. Detects a band of approximately 24 kDa (predicted molecular weight: 24 kDa).
IHC-P: Use a concentration of 10 µg/ml. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.
Complexes with metalloproteinases (such as collagenases) and irreversibly inactivates them by binding to their catalytic zinc cofactor. Known to act on MMP-1, MMP-2, MMP-3, MMP-7, MMP-8, MMP-9, MMP-10, MMP-13, MMP-14, MMP-15, MMP-16 and MMP-19.
Belongs to the protease inhibitor I35 (TIMP) family.
Contains 1 NTR domain.
The activity of TIMP2 is dependent on the presence of disulfide bonds.
Secreted.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections)

IHC image of TIMP2 staining in human pancreatic cancer FFPE section, performed on a BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab109708, 10µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX

Western blot

Anti-TIMP2 antibody [3A4] (ab109708) at 5 µg/ml + Recombinant TIMP2 at 0.1 µg
Secondary
Goat polyclonal Secondary Antibody to Mouse IgG - H&L (HRP), pre-adsorbed (ab97040) at 1/5000 dilution
developed using the ECL technique
Performed under reducing conditions.
Observed band size : 24 kDa (why is the actual band size different from the predicted?)
Exposure time : 20 minutes
