Cooperates with LY96 to mediate the innate immune response to bacterial lipoproteins and other microbial cell wall components. Cooperates with TLR1 to mediate the innate immune response to bacterial lipoproteins or lipopeptides. Acts via MYD88 and TRAF6, leading to NF-kappa-B activation, cytokine secretion and the inflammatory response. May also promote apoptosis in response to lipoproteins. Recognizes mycoplasmal macrophage-activating lipopeptide-2kD (MALP-2), soluble tuberculosis factor (STF), phenol-soluble modulin (PSM) and B.burgdorferi outer surface protein A lipoprotein (OspA-L) cooperatively with TLR6.
Highly expressed in peripheral blood leukocytes, in particular in monocytes, in bone marrow, lymph node and in spleen. Also detected in lung and in fetal liver. Levels are low in other tissues.
Belongs to the Toll-like receptor family.
Contains 14 LRR (leucine-rich) repeats.
Contains 1 TIR domain.
Glycosylation of Asn-442 is critical for secretion of the N-terminal ectodomain of TLR2.

ab24192 staining TLR2 in Human spleen tissue sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections). Tissue was fixed with formaldehyde; antigen retrieval was by heat mediation with a citrate (pH 6.0) or EDTA (pH 9.0) buffer. Samples were incubated with primary antibody (1/400 in antibody diluent + background reducers) for 20 minutes at 25°C. An undiluted HRP-conjugated Goat polymer was used as the secondary antibody.

ab24192 staining TLR2 in human skin tissue by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections).Tissue was fixed in paraformaldehyde and a heat meditated antigen retrieval step was performed using citrate buffer pH 6.0. Samples were then blocked using 4% BSA for 30 minutes at 25°C and then incubated with ab24192 at 2µg/ml for 14 hours at 4°C. The secondary used was a biotin conjugated goat anti-rabbit polyclonal used at 10µg/ml.The enzyme for detection was alkaline phosphatase and additonal counterstaining was performed with Hematoxylin.
