Mouse monoclonal [TRA-1-81] to TRA-1-81 - Embryonic Stem Cell Marker
This antibody reacts with TRA-1-81 antigen that is expressed upon the surface of human teratocarcinoma stem cells (EC), human embryonic germ cells (EG) and human embryonic stem cells (ES). No immunoreactivity is seen with murine EC, EG or ES cells.Both the TRA-1-81 and TRA-1-60 monoclonal antibodies recognize antigens that are associated with a pericellular matrix proteoglycan. TRA-1-60 reacts with a sialidase-sensitive epitope whilst TRA-1-81 reacts with an unknown epitope of the same molecule. The epitopes for both TRA-1-81 and TRA-1-60 are not confined to human EC cells, but are also found on several neoplastic and normal tissues, though in a different pattern for each antibody. TRA-1-81 immunoprecipitates a 420kD and a 250kD protein and TRA-1-60 immunoprecipitate a 235kD and a 410kD protein from lysates of human EC cells.

The Podocalyxin gene encodes a member of the sialomucin protein family, and the encoded protein was originally identified as an important component of glomerular podocytes. Other biological activities of the encoded protein include: playing a role in hematopoetic cell differentiation, binding in a membrane protein complex with Na+/H+ exchanger regulatory factor to intracellular cytoskeletal elements and being expressed in vascular endothelium cells and binding to L-selectin.

ab16289 at 1/100 staining human embryonic stem cells by ICC/IF. The cells were paraformaldehyde fixed and blocked with 5% goat serum prior to incubation with the antibody for 1 hour. An Alexa-Fluor 488 conjugated Goat anti-mouse IgM was used as the secondary antibody.
DAPI staining is shown in the top panel while TRA-1-81 staining with ab16289 is shown in the lower panel.

Cells were paraformaldehyde fixed and permeabilized in Triton prior to blocking in 10% serum for 1 hour at 24°C. The primary antibody was diluted 1/500 and incubated with the sample for 1 hour at 37°C. An Alexafluor 647 goat polyclonal to mouse IgM, diluted 1/200, was used as the secondary.
