Mouse monoclonal [19A449] to Tankyrase

Specificity

Ab13587 recognizes tankyrase 1. It may also recognize tankyrase 2 due to the high homology between tankyrase 1 and 2. However, the epitope has not been mapped to determine if it is shared between tankyrase 1 and 2. Tankyrase 1 is a 1327 amino acid protein; Genbank accession number AAC79841. The full-length human tankyrase 1 used as immunogen to generate ab13587 contains the amino acid sequence listed in GenBank no. AAC79841. Tankyrase 2 is a 1126 amino acid protein; Genbank accession number NP_079511. Tankyrase 1 produces, by alternative splicing, 8 different transcripts putatively encoding 8 different isoforms. Tankyrase 2 putatively encodes 8 different isoforms. Therefore the molecular weight of tankyrase observed by western blot with the antibody may vary depending on the isoform expressed and recognized by the antibody. Users are encourage to refer to NCBI AceView http://www.ncbi.nlm.nih.gov/IEB/Research/Acembly/ for more information about tankyrase isoforms. The epitope recognized by this antibody has not been mapped. It would be difficult to distinguish between tankyrase 1 and tankyrase 2 with ab13587 by western blot alone since certain isoforms may migrate at similar molecular weight. For example, a 1094 amino acid form of tankyrase 1 is listed in GenBank (no. AAC79842.1), which is similar in size to the amino acid size to tankyrase 2.

During interphase, a small fraction of TNKS is found in the nucleus, associated with TERF1.

ab13587 at 1/150 staining mouse small intestine tissue sections by IHC-P. The tissue was paraformaldeyde fixed and a heat mediated antigen retrieval step was performed in citrate buffer before incubation with the antibody for 45 minutes. An HRP conjugated goat anti-mouse antibody was used as the secondary.

ICC/IF image of ab13587 stained Mcf7 cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab13587, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-mouse IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

Overlay histogram showing HEK293 cells stained with ab13587 (red line). The cells were fixed with 100% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab13587, 1/100 dilution) for 30 min at 22°C. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22°C. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a positive signal in HeLa cells fixed with 4% paraformaldehyde (10 min)/permeabilized in 0.1% PBS-Tween used under the same conditions.
