thank you so much for your advice, just to be sure, these recommendations are based on:

http://www.ncbi.nlm.nih.gov/nuccore/AF018167 ?

Sorry for being long winded, the customer is rather stringent on her choices.

Have a nice day!

Many Thanks and Best Regards,

Deng Kai Jun

ANSWER:

Thank you for contacting us again.

We usually, if not mentioned on the datasheet, use the protein sequencesas described in SwissProt (ID#O14746). This database, however, has a link to NCBI Sequence databases as a cross- reference.

I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.

One of our customers is looking for an antibody against hEST, she's
studying the following sequence:
http://www.ncbi.nlm.nih.gov/nuccore/AF018167
She would use this on human samples for IF. our customer is working with cells.

ANSWER:

Thank you for contacting us.

I would like to suggestthe followingtwo antibodies which are suitable for your customer's purpose:

http://www.abcam.com/index.html?datasheet=5181 (or use the following: http://www.abcam.com/index.html?datasheet=5181).

ab5181 Anti-Telomerase reverse transcriptase antibody [2C4]has one reference with the use in ICC.

http://www.abcam.com/index.html?datasheet=32020 (or use the following: http://www.abcam.com/index.html?datasheet=32020).

ab32020 Anti-Telomerase reverse transcriptase antibody [Y182]has two customer raitings and one reference for the use in ICC.

Question 3

Tuesday 10-April-2012

Hi Technical,
Kindly provide a recommended protocol for ab5181 (anti-Telomerase) using IHC-P applications i.e. dilution factor, pre-treatment method.
Also, please advise the appropriate positive control to be used.
Thanking you in advance.

ANSWER:

Thank you for contacting us.

There isn’t a specific protocol for performing IHC-P using this antibody. According to the datasheet the antibody concentration is assay dependant, and optimisation is recommended by using different antibody dilutions in order to determine the most suitable for your particular conditions.

You can find on the dustsheet an Abreview submitted by another user of ab5181 in IHC-P that may serve as guidance for your assay. As a positive control pancreatic carcinoma and normal pancreas (human) are suggested.

The following links will lead you to the IHC-P protocols from our Website, with useful tips and explanations that may be very useful and could serve as a recommended protocol for your staining:

Question 4

Friday 30-December-2011

Thank you for your email. Our customer ordered previously 4 vials and had optimized condition using this antibody for almost one year. After this, he purchased 10 vials more but product type changed suddenly, he is very embarrased now. Hegot 100ul beforebut now its volume is 500ul. This antibody cannot check the concentration but is there any standard for this packing size? Please let him know if you can suggest or recommend about concentration information. And, Happy new year!

ANSWER:

Thank you for your reply and I understand your customer's concern. We were supplying this antibody from an outside lab and were selling it at 100ul and it was unpurified so the concentration was not determined. Now we currently make this clone in house and are selling it at 500ul, but still it is unpurified so we do not know the concentration. A rough estimate of the protein concentration in a tissue culture supernatant is between 1-3 mg/ml. I hope this information helps. Please contact us with any other questions.

Question 5

Thursday 29-December-2011

What is the concentration?

ANSWER:

Thank you for your reply. The concentration for all lots of this antibody (GR59361-1 and GR22704-2) have not been determined because it is a concentrated tissue culture supernatant. I apologize for the misinformation on the datasheet. I am having our Datasheets Team update the concentration on our website for this product to read "concentration not determined," but this usually takes a few days to go through. Thank you for your understanding. Please let me know if you have any other questions.
