Specificity
|
MTn-12 (Aufderheide et al. 1988) reacts specifically with mouse tenascin. In supernatants of cultured mouse fibroblasts, the antibody precipitates two polypeptides of ~210 and ~260 kDa. It also recognizes these tenascin chains and the 230 kDa endoglycosidase F-digested fragment in immunoblotting of tissue extracts and fibroblast lysates under reducing conditions (Aufderheide et al. 1988, Weller et al. 1991).
In ELISA, it was shown that the antibody reacts specifically against mouse tenascin, but not against fibronectin and collagen type I.
Immunoprecipitation data suggests that molecules of the expected size for tenascin-C are detected by MTn-12, and not other tenascins. One can expect several bands in blots or immunoprecipitations but this is because the antibody can detect several splice variants of tenascin C. Nevertheless, the mapping of the epitope and the other data cannot entirely exclude the possibility that it reacts with other tenascins. This would have to be tested directly with purified tenascins.

IF: 1/200 for staining of unfixed, frozen tissue sections of mouse intestine (also see reference Wirl et al). IHC-Fr: Use at an assay dependant dilution. IHC-FoFr: 1/100. IP: Use at a concentration of 50 µg/ml in 200 µl Tris-buffered saline (TBS)(see reference Wirl et al). WB: 1/20 (see references Wirl et al and Weller et al). Block with 0.1% gelatine in PBS (see Wirl et al).
Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Tenascin, also known as hexabrachion and cytotactin, is an extracellular matrix protein with a spatially and temporally restricted tissue distribution. It is a hexameric, multidomain protein with disulfide linked subunits of 190 to 240 kD, originally characterized as 'myotendinous antigen.' In the embryo it is present in dense mesenchyme surrounding developing epithelia and in developing cartilage and bone. In the adult, tenascin remains present in tendons and myotendinous junctions in the perichondrium and periosteum, as well as in smooth muscle.

ab6346 staining Tenascin C in mouse brain tissue section by Immunohistochemistry (PFA perfusion fixed Frozen sections). Tissue samples were fixed with paraformaldehyde and blocked with 10% serum for 30 minutes at 250C. The sample was incubated with primary antibody (1/100) for 16 hours at 250C in 10% NGS in PBS + 0.1% TX100.
