We have ordered many antibodies from Abcam because we think that they work pretty good. We are performing experiments on extracellular matrices and antibodies against type I collagen, typa III collagen, type IV collagenn works greatly. However i have some concerns about hhe antibody Mouse monoclonal to Thrombospondin (ab3131). We've bought all this antibodies last years but after repeated assays i think that the thrombospondin one has some limits. We have used it in immunofluorescence on frosen sections, and effectively it stains something. However, after different western blotting in reducing and non reducing conditions, we noticed that it does not recognize a band of 450 KDa as stated in the datasheet but always bands lower than 60 KDa. Lysates were from murine megakaryocyte (well-documented producers of this protein). In the last year no abreviews have been posted on its internet page and this antibody has not yet been referenced in any publicautions. Could you please give some explanation?It seems that the clone A6.1 works better, is it possible to get a replacement with this clone?

Concerning molecular weight: On WB this antibody detects ~450 kDa non-reduced TSH. What band or bands are detected under reduced conditions?

A customer wants to know if ab1823 and ab3131 are for Thrombospondin 1, 2 or both?

Thank you for your enquiry.

Following contact with the source of this antiserum I have determined that ab3131 inhibits anti-angiogenic activity of TSP-1 by inhibiting its binding to TSP-receptor / CD36. I have been informed that it has specificity towards TSP1.

ab1823 does cross react against TSP type 2.
