Recommended dilutions Flow Cytometry / Immunofluorescence: assay dependent. Western Blotting (Non-reduced only): use at a concentration of 1 - 2µg/ml for 2hrs at RT. Not tested in other applications.
Optimal dilutions / concentrations should be determined by the end user.
By Western blot, this antibody detects a band of ~ 450 kDa, which corresponds to the predicted molecular weight of Thrombospondin.
This antibody blocks anti-angiogenic activity of TSP 1 by inhibiting its binding to TSP Receptor / CD36.
This antibody inhibits the adhesion of human melanoma G361 cells, keratinocytes, squamous carcinoma cells, and rat smooth muscle cells to TSP 1.
It does not inhibit aggregation of thrombin-induced platelets.

Thrombospondin is a regulator of many biological processes including cell growth, adhesion, migration, platelet aggregation, and fibrin deposition and lysis. It interacts with a number of plasma proteins such as fibrinogen and plasminogen and co-polymerizes with fibrin in clot formation. Thrombospondin also has multiple binding sites that interact with molecules such as fibronectin, collagens, laminin and heparan sulphate proteoglycans as well as binding growth factors such as TGF-beta1. Thrombospondin exerts an anti-adhesive effect which leads to cell rounding and detachment. Thrombospondin is also implicated in a key role in brain development and synapse regulation.
