I'm interested in your anti-Titin. You can use this antibody for western blot but I don't have a protocol for a protein that big.The standard protocols are not designed for such high weight molecules. Do you have a protocol for this? If you have can you send it to me. Thanks

Thank you for your enquiry regarding separation on WB of large proteins.

I would recommend using a 5% 1cm thick gel and to change the ratio of bis-acrylamide to mono-acrylamide from the classical 37:1 ratio to a 49:1 ratio.

For the transfer I recommend adding a little SDS to the transfer buffer to help.

By DD-PCR,we found that titin can be induced to express on trophoblasts.We blasted the DD-PCR product(92 bp) to geneBank,they are the same as the 404th to 495th bases of novex mRNA.now I want to detect the DD-PCR products in protein level Can I use the antibody of titin(ab7034)? I hope Ican get your kindly help soon.Thanks a lot.

ANSWER:

 

Yes, you can use this antibody for Western blotting on human tissue samples.

I am afraid that we don't know the size that Titin runs on a Western blot. I do know that the antibody detects a doublet though......we don't have a Western blot picture using this antibody.

As we rely on feedback from customers and other sources for a lot of our data, we sometimes don't get all the information we want, but we feel that limited information is better than none. We would rather present you with all the information we have and let the customer decide.

I don't have a specific protocol for lysate preparation for use with this antibody, I can only suggest you check the literature. I know that as titin has a very high molecular weight you will find it difficult to use standard protocols. I did see an interesting looking paper on this recently (Electrophoresis. 2003 Jun;24(11):1695-702.) I believe you should find some useful advice in there.

I have few questions about the titin - ab7034 antibody, you carry. I would like to get your conformation that is reacts with the human cardiac tissue! Do you know the epitope of the antibody? Approximately. Is it C-termin. or N-termin? For Western blotting what dilution is needed? Would you be so kind to make a bid for the above mentioned product?

Yes, this antibody should cross react with human cardiac muscle. We have no information on the location of the epitope.
