Thank you for your inquiry. I heard back from the supplying laboratory that they do not determine the precise concentration of IgG for ab84478. This product was prepared to provide specific IHC staining within the recommended range of dilutions. The concentration of IgG is less than or equal to 1 mg/ml. For your purposes where you wish to run a normal rabbit IgG as a control, assuming 1 mg/ml for the concentration of IgG in the IHC antibody is reasonable. I hope this information helps. ab84478, at a 1/250 dilution, staining UACA in formalin fixed, paraffin embedded human breast carcinoma tissue section by Immunohistochemistry.
Detection: DAB staining
ICC/IF image of ab84478 stained HeLa cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab84478, 10µg/ml) overnight at +4ºC. The secondary antibody (green) was ab96899, DyLight® 488 goat anti-rabbit IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
