Vav proteins are members of the Dbl-family of GEFs (Guanine Nucleotide Exchange Factors) for the Rho/Rac-family GTPases which activate pathways leading to actin cytoskeletal rearrangements and transcriptional alterations. Vav proteins play pivotal roles in cytoskeletal re-organization in processes such as cell division and adhesion, phagocytosis, and platelet activation. At least three Vav isoforms have been identified; Vav1, Vav2 and Vav3. VAV3 is expressed in many tissues and participates in signal transduction processes. It is involved in the activation of several members of the Rho family. This activity, when deregulated, leads to marked cytoskeletal changes and to alterations in the process of cell division. Vav3 promotes PLC and PI3 Kinase activation, thus mediating Ca2+-dependent signalling.

VAV3 antibody (ab21208) at 1 µg/ml + Human Bone Marrow lysate (35µg protein in RIPA buffer).
Predicted band size : 90 kDa Observed band size : 98 kDa (why is the actual band size different from the predicted?)
Primary incubation was 1 hour. Detected by chemiluminescence.
